Abstract:RNA editing is a crucial process in post-transcriptional modification. Among various types of RNA editing, A-to-I (adenosine-to-inosine) RNA editing is the most predominant and common one, which is catalyzed by adenosine deaminases acting on RNA (ADARs). Notably, ADAR1 is the most important isoform in the ADAR family. In recent years, the role of ADAR1 in multiple biological processes has attracted extensive attention, particularly in the field of innate immunity. Inflammation is a key component of innate immunity. ADAR1 participates in the regulation of inflammatory responses by editing immunity-related genes, affecting the recognition of double-stranded RNA (dsRNA) by immune sensors, and modulating the activation of inflammatory signaling pathways. The expression and activity of ADAR1 are regulated by a variety of signaling pathways, which play critical roles in the inflammatory process. Furthermore, abnormal ADAR1 activity has been observed in a range of diseases, including acute inflammatory diseases, autoimmune diseases, and viral infections. This review aims to summarize the regulatory mechanisms of ADAR1 in inflammatory responses and the research progress on diseases caused by abnormal ADAR1 activity. This review, based on the overall understanding formed from the latest research, aims to summarize the regulatory mechanism of ADAR1 on inflammatory responses and to review the latest research progress on diseases caused by abnormal ADAR1 activity.
