目的 探究长链非编码RNA(lncRNA)LUCAT1对人胰腺癌细胞MIA PaCa-2生物学行为的影响，探讨LUCAT1在胰腺癌恶性进展中的潜在作用。方法 在线数据库GEPIA分析LUCAT1在胰腺癌中的突变及表达情况；q-PCR实验检测对比人胰腺导管细胞HPNE及人胰腺癌细胞中LUCAT1的表达水平；FISH检测人胰腺癌组织中LUCAT1表达及分布。CCK-8实验和Transwell实验检测LUCAT1对MIA PaCa-2细胞增殖、凋亡、耐药及迁移能力的影响。基因集合富集分析比对LUCAT1参与的相关信号通路，Western blot实验验证蛋白质表达水平。结果 GEPIA分析结果显示，LUCAT1在人胰腺癌组织中表达水平上调；q-PCR实验结果显示，人胰腺癌细胞中LUCAT1高表达，差异有统计学意义(P<0.05)。敲低及过表达LUCAT1可影响胰腺癌细胞增殖、凋亡、吉西他滨耐药性以及迁移和侵袭能力(P<0.05)。在胰腺癌中LUCAT1影响p-Akt表达水平，并在Akt抑制剂MK-2206处理后相关功能被抑制。结论 LUCAT1通过PI3K-Akt信号通路调控胰腺癌细胞MIA PaCa-2恶性进展。
